The recommendation seems to say when putting in a spline is to use carpenters glue to glue the spline in and then followup that up with a cleat or staple. My question is do you need to let the glue dry before nailing? I searched the site and couldn't find a clear cut answer to that.

No need to wait. In fact, better not to. Then you have no worries about excess dried glue preventing boards from going together.

When nailing the spline in, have a small scrap with the groove. Place it next to the nailer or the nailer next to it with the groove engaged on the spline, and nail the spline. This keeps the spline straight and at the right spot.
